T7 RNA polymerase (RNApoly)

T7 RNA Polymerase (T7 RNA polymerase) is a highly DNA-dependent 5'→3' RNA polymerase that recognizes the T7 promoter sequence. T7 RNA Polymerase catalyzes the incorporation of NTP downstream of the single or double-stranded DNA T7 promoter to synthesize RNA that complements the template DNA downstream of the T7 promoter.Despite its simple structure in comparison to polymeric prokaryotic and eukaryotic RNA polymerases, T7 RNA polymerase (Pol) performs highly specific promoter recognition and all other steps in the transcription process in the absence of any other protein factors. These and other properties make T7(and similar SP6 and T3) RNA polymerases particularly suitable for the study of transcriptional mechanisms. In recombinant DNA technology, T7 RNA polymerase is used as a powerful tool to drive high level expression of cloned genes and/or to prepare a large number of defined RNA transcripts in vitro.
