Zinc Finger Protein 175 (ZNF175)
OTK18 contains an N-terminal KRAB box and a C-terminal zinc finger region. Confocal microscopy localized OTK18 to both cytosol and nucleus.The 3,754-nucleotide clone encodes a predicted 711-amino acid protein. Since the protein contains 13 contiguous zinc finger structures of the C2H2 type, it was expected to function as a DNA-binding multifinger protein. A 4.3-kb OTK18 transcript was expressed in all adult human tissues examined by Southern analysis.both the KRAB and zinc finger regions of OTK18 were required for its transcriptional suppressor function. OTK18 was induced in macrophages following HIV-1 infection and restricted viral infection. OTK18-mediated suppression of HIV-1 replication appeared to involve suppression of HIV-1 Tat-induced viral long-terminal repeat promoter activity.
