Zinc Finger Protein 333 (ZNF333)
ZNF333 recognized the specific DNA core binding sequence ATAAT. ZNF333 repressed transcription of promoters containing the ATAAT sequence in a luciferase reporter assay.
The deduced 665-amino acid protein has a calculated molecular mass of 75.5 kD and shares 44% amino acid identity with ZNF157. ZNF333 has 10 C2H2 zinc finger motifs at the C terminus and 2 N-terminal KRAB domains, both containing KRAB A and KRAB B domains. EST database analysis and RT-PCR identified a splice variant with 174 amino acids, only 1 KRAB domain, and lacking zinc finger motifs. Northern blot analysis of human tissues detected a 4.6-kb transcript with strong expression in heart, moderate expression in liver, skeletal muscle, kidney, and pancreas, and low expression in placenta.
