TBC1 Domain Family, Member 2 (TBC1D2)
Using the SEREX (serological identification of antigens by recombinant expression cloning) technique, Zhou et al. (2002) identified an immunogenic tumor antigen from prostate cancer cell lines. The corresponding TBC1D2 gene, which the authors called PARIS-1 (prostate antigen recognized and identified by SEREX), encodes a 917-amino acid protein with a predicted molecular mass of 104 kD. In vitro translation of TBC1D2 revealed a doublet of approximately 118 kD, suggesting posttranslational modification. TBC1D2 contains a TBC domain, which is conserved in RAB GTPase-activating proteins, and numerous glycosylation and phosphorylation sites. Northern blot analysis identified a 3.3-kb TBC1D2 transcript with ubiquitous but variable expression levels in a variety of normal and cancer cell lines.
