Pleckstrin Homology Domain Containing Family A, Member 7 (PLEKHA7)
The deduced 1,122-amino acid protein contains an N-terminal WW domain, followed by a pleckstrin (PLEK) homology region and a coiled-coil domain. Western blot analysis of Caco2 cells detected major and minor PLEKHA7 proteins with apparent molecular masses of 135 and 145 kD, respectively. Immunofluorescence analysis showed that PLEKHA7 localized to cell-cell junctions in Caco2 cells and colocalized with zona adherens proteins.
Protein pull-down assays revealed that PLEKHA7 interacted with the junctional protein Nezha. Depletion of PLEKHA7 reduced the amount of Nezha associated with cell junctions, and depletion of Nezha reciprocally altered the pattern of PLEKHA7 distribution.
