Multiple EGF Like Domains Protein 10 (MEGF10)
Most MEGF10 was restricted to the basal surface of transfected cells, and it was not expressed at the cell edge or in lamellipodia. MEGF10 bound the cells tightly to coated glass, which resulted in a flattened phenotype, restricted cell motility, and hindered cell division. Time-lapse fluorescence microscopy suggested that MEGF10 monomers bound the coated glass surface and that additional MEGF10 molecules accumulated into multimers that ultimately formed a complex network of proteins in an irregular mosaic pattern, binding the cell membrane to the substrate. EMI domain and the 17 atypical EGF-like domains of MEGF10 were required for the mosaic pattern of expression. Coculture of MEGF10-expressing cells with apoptotic cells revealed that MEGF10 accumulated around the contact region during engulfment of apoptotic cells.
