PDS5, Regulator Of Cohesion Maintenance Homolog A (PDS5A)
The deduced 1,297-amino acid protein had a calculated molecular mass of 146 kD. PDS5A contains an N-terminal RhoGEF domain, a leucine zipper domain, an N-adaptin domain, a SKP1 domain, 3 PEST sequences, 6 nuclear localization signals, and 2 sites for tyrosine phosphorylation. Northern blot analysis detected transcripts of about 7 and 8 kb with high expression in adult skeletal muscle, heart, placenta, liver, and kidney. Western blot analysis detected PDS5A at an apparent molecular mass of 150 kD in a human lung carcinoma cell line and in human colon, lung, ovary, breast, and kidney tissues. An additional band of about 65 kD was also detected in most of the 22 normal kidney tissues examined. Immunohistochemical analysis of breast cancer cells showed PDS5A localized to the nucleus.
