REST Corepressor 1 (RCOR1)
The RCOR gene encodes a functional corepressor required for regulation of neural-specific gene expression. The predicted COREST primary structure contains 2 SANT (SW13/ADA2/NCoR/TFIIIB) domains separated by 191 amino acids. A similar module exists in NCOR/SMRT and in the transcription factor Adf1, suggesting that there is a conserved functional role for this arrangement of SANT domains. Drosophila and human COREST predicted proteins are 84% identical over a span of 114 amino acids including a SANT domain that is nearly identical in the 2 proteins. COREST is ubiquitously expressed in adult human tissues. COREST amino acids 109 through 293 interacted strongly with the C-terminal half of REST, as well as with a smaller C-terminal REST fragment containing a zinc finger motif that mediates repressor activity.
