Pleckstrin Homology Domain Containing Family M, Member 1 (PLEKHM1)
PLEKHM1 may have critical function in vesicular transport in osteoclasts. The deduced 926-amino acid protein has a calculated molecular mass of 103.2 kD and contains an N-terminal leucine zipper motif and 2 central pleckstrin homology (PH) domains. Northern blot analysis detected a major 5.7-kb transcript in all tissues and cancer cell lines examined, with highest expression in thymus, prostate, spleen, and peripheral blood lymphocytes. A transcript of 8.7 kb was also detected in thymus, and transcripts of 3.8 and 3.2 kb were also detected in spleen, thymus, and testis.RT-PCR detected moderate expression in placenta, liver, and ovary and low expression in brain, lung, kidney, thymus, prostate, and testis. No expression was detected in heart, skeletal muscle, pancreas, spleen, and small intestine.
