Pleckstrin Homology Like Domain Family B, Member 1 (PHLDB1)
PHLDB1 contains at least 1,234 amino acids and shares a high degree of similarity with rat Ll5. RT-PCR detected PHLDB1 in all tissues examined, with highest expression in ovary, brain, lung, and kidney. The full-length 1,377-amino acid protein contains an N-terminal phosphorylation-dependent forkhead-associated (FHA) protein interaction domain, followed by a bipartite nuclear localization signal, a central chromosome segregation ATPase (SMC) domain, and a C-terminal pleckstrin (PLEK) homology (PH) domain. EST database analysis suggested that variant-1 was the major transcript.isolated C-terminal PH domain of LL5A interacted with all phosphoinositides examined, with highest affinity for phosphatidylinositol 3,4,5-trisphosphate (PtdIns(3,4,5)P3). It did not interact with PtdIns or with any other phospholipid examined.
