COMM Domain Containing Protein 1 (COMMD1)
COMMD1 is a regulator of copper homeostasis, sodium uptake, and NF-kappa-B signaling. The predicted human, mouse, and canine proteins share 86 to 87% amino acid identity. Northern blot analysis of human tissues using a canine Murr1 probe revealed highest expression in liver, with lower expression in heart, skeletal muscle, kidney, and placenta.This inhibition was mediated in part through its ability to inhibit basal and cytokine-stimulated nuclear factor NF-kappa-B activity. Knockdown of MURR1 increased NF-kappa-B activity and decreased IKBA concentrations by facilitating phospho-IKBA degradation by the proteasome. MURR1 was detected in CD4+ T cells, and RNA-mediated interference of MURR1 in primary resting CD4+ lymphocytes increased HIV-1 replication.
