Fiz1 encodes a 500-amino acid protein with a predicted molecular weight of 52kD. Sequence analysis revealed the presence of 11 C2H2-type zinc fingers. Northern blot analysis of RNA from various mouse cell lines and tissues detected ubiquitous expression of a 2.6-kb transcript.NRL regulates the expression of several rod-specific genes, and missense mutations in the human NRL gene are associated with autosomal dominant retinitis pigmentosa.
Fiz1 binds to the catalytic domain of Flt3 but not to the structurally related receptor tyrosine kinases Kit, Fms, or platelet-derived growth factor receptor (PDGFRA). This association is independent of kinase activity. Anti-Fiz1 antibodies recognized a 60-kD protein, which was localized in the nucleus as well as in the cytoplasm.
