HES5 regulates brain development process. HES5 expression significantly higher in squamous cervical carcinoma than in CIN as well as higher in CIN than normal cervical epithelia. Human HES5 gene binds to Notch receptor and expression of HES5 decreases during cartilage differentiation.

Hes5 contains a basic helix-loop-helix (bHLH) domain that shares 96% identity with the bHLH region of rat Hes5. In situ hybridization showed that Hes5 was expressed at high levels throughout the developing mouse central nervous system. deduced 166-amino acid protein contains an N-terminal bHLH domain, followed by an orange domain, a proline-rich domain, and a C-terminal WRPW motif. HES5 ESTs were found in fetal heart and brain tumor cDNA libraries.
