By sequencing clones obtained from a size-fractionated brain cDNA library, Hirosawa et al. (1999) cloned CASKIN2, which they designated KIAA1139. The deduced protein contains motifs associated with cell signaling. RT-PCR ELISA detected ubiquitous expression of CASKIN2, with highest levels in fetal and adult liver.
Tabuchi et al. (2002) found that Caskin proteins are highly conserved evolutionarily. Each has a multidomain structure in which the N terminus has 6 ankyrin repeats, a single SH3 domain, and 2 SAM domains, and the C terminus contains a long proline-rich sequence and a short conserved domain. Human CASKIN2 encodes a deduced 1,202-amino acid protein.
