CINP promoted cell viability in response to replication stress and was required for efficient ATR-dependent signaling after DNA damage. Silencing of CINP also increased the percentage of cells escaping the G2 checkpoint following ionizing or ultraviolet irradiation.CINP also interacted with the DNA-replication licensing complex protein MCM5, but it did not interact with with RB (RB1) or other CDK2-interacting proteins. In vitro coimmunoprecipitation assays identified CINP as a component of cyclin E (CCNE1)/CDK2 and cyclin A (CCNA1)/CDK2 complexes. CINP also interacted with CDC7 (CDC7L1) and was phosphorylated by CDC7, but not by CDK2. CINP bound to chromatin in a replication-dependent manner and associated with the origin recognition complex protein ORC2 (ORC2L).
