Nuclear Casein Kinase And Cyclin Dependent Kinase Substrate 1 (NUCKS1)
The deduced human and rat proteins contain 243 amino acids and share 95.1% sequence identity. By immunoblotting of intracellular fractions and fluorescence microscopy of HEK-293 cells expressing NUCKS1-GFP, Ostvold et al. (2001) determined that NUCKS1 is a nuclear protein. Grundt et al. (2004) found NUCKS1 orthologs in several species from zebrafish to mouse, all of which encoded highly conserved proteins compared to human NUCKS1. No orthologs were found in insects or yeast, suggesting that NUCKS1 is a vertebrate gene. The conserved regions of NUCKS1 contain several consensus phosphorylation sites for casein kinase II (CK2) and cyclin-dependent kinases, 2 putative nuclear localization signals, and the basic DNA-binding domain. The N terminus is extremely well conserved.
