PH Domain And Leucine Rich Repeat Protein Phosphatase 1 (PHLPP1)
The deduced 1,696-amino acid protein has an N-terminal pleckstrin homology (PH) domain, followed by 18 leucine-rich repeats (LRR), a leucine zipper, a protein phosphatase-2C (PP2C)-like domain, and a C-terminal polyglutamine stretch. Western blot analysis of several rat tissues detected Scop in cerebrum, cerebellum, and testis but not in any other tissue examined. The C-terminal region also has a glutamine-rich region with a 6-residue polyglutamine stretch. EST and microarray analyses indicated that PHLPP is ubiquitously expressed, with highest levels in brain. Western blot analysis detected 2 endogenous PHLPP proteins in human embryonic kidney cell lysates: a major protein with an apparent molecular mass of about 135 kD, and minor protein with an apparent molecular mass of about 140 kD.
