Thioesterase Superfamily, Member 4 (THEM4)
CTMP negatively regulates PKB activity by inhibiting phosphorylation of these PKB residues. The deduced 240-amino acid CTMP protein is 79% identical to the mouse protein. RT-PCR analysis detected CTMP expression predominantly in skeletal muscle, testis, uterus, brain, and kidney, with lower levels in heart, liver, and lung. The presence of multiple transcripts suggested that CTMP undergoes alternative splicing. Western blot analysis showed expression of a 22- to 26-kD protein in different cell lines. Immunoprecipitation, immunoblot analysis, and immunofluorescence microscopy demonstrated predominant expression in the plasma membrane, where CTMP colocalizes with PKB. Cinemicroscopy illustrated localization of CTMP to the leading edge of moving cells.
