Diacylglycerol lipase is a key enzyme in the biosynthesis of the endocannabinoid 2-arachidonoylglycerol. It catalyzes the hydrolysis of diacylglycerol releasing a free fatty acid and monoacylglycerol.Two separate genes encoding DGL enzymes have been cloned, termed DGLα (DAGLA) and DGLβ (DAGLB), that share 33% sequence identity.
The enzyme has been described to be inhibited selectively by two agents, RHC80267 and tetrahydrolipstatin.Catalyzes the hydrolysis of diacylglycerol (DAG) to 2-arachidonoyl-glycerol (2-AG), the most abundant endocannabinoid in tissues. Required for axonal growth during development and for retrograde synaptic signaling at mature synapses.
