Glucagon-like Peptide 1--a Pioneer of Diabetes Drugs

Diabetes (diabetes mellitus) is a metabolic disease that is characterized by hyperglycemia. Due to long-term high blood sugar in diabetic patients, this syndrome is prone to cause a variety of complications. Then leading to chronic damage and dysfunction in multiple organizations, especially in the eye, kidney, heart, blood vessel, nerve and other tissues. Therefore, treatment and control of diabetes has become one of the hot spots of medical field.

As increasing studies on the mechanism of diabetes in the pathogenesis, an amazing molecule that can control blood sugar slowly opened its mysterious veil until 1987. Researchers have found that gastrointestinal endocrine cells will produce a protein that shares a high homology with pre-pro-glucagon, this molecule is capable of promoting insulin secretion by stimulating pancreatic β cell proliferation, preventing apoptosis of pancreatic β cells, inhibiting of glucagon’s release, etc. which will control blood sugar levels. It is called GLP-1 (glucagon-like peptide-1, glucagon-like peptide-1).

Glucagon-like peptide-1 (GLP-1) is mainly secreted by L-cells of the intestinal mucosa, it is expressed by pro-glucagon gene and cleaved into its carboxy terminal peptide sequence, i.e., GLP-1, by pro-hormone converting enzyme (PC1). GLP-1 has two kinds of biologically active forms, namely GLP-1 (7-37) and GLP-1 (7-36) amide. In addition to GLP-1, sequences of many peptides are very similar to that of GLP-1 in the body (Figure 1). Meanwhile, because of the immunogenicity of GLP1 is too week to produce its antibody. Thus, it is very difficult for researchers to detect the level of GLP-1 in vivo.

GLP-1--a Pioneer of Diabetes Drugs

In order to enhance the immunogenicity of GLP1 and avoid non-specific recognition of GLP-1 antibody, Cloud-Clone Corp. synthesized human glucagon-like peptide-1 (GLP-1) polypeptide (Cat No: CPA804Hu01) and coupled it with OVA (glucagon-like peptide 1 (GLP1)-OVA conjugate, Cat No: CPA804Hu21), this optimization not only avoided non-specific peptide identification between this antibody and other irrelevant peptides, but also greatly improved the immunogenicity of GLP1 polypeptide. Besides, Cloud-Clone Corp. successfully obtained polyclonal antibody to human glucagon-like peptide 1 (GLP1) (Cat No: PAA804Hu01) with high specificity and high titer. This antibody can specifically recognize human GLP-1 peptide, amd no significant cross-reactivity or interference between GLP1 and analogues was observed by Western blotting test. In addition, Cloud-Clone Corp. has also successfully developed ELISA Kit for Glucagon Like Peptide 1 (Cat No: CEA804Hu). The kit can quantitatively detect the level of GLP-1 among a variety of human samples, including serum, plasma, tissue homogenate, cell lysates, cell culture supernatants and other biological fluids.

In addition to the products above, Cloud-Clone Corp. had also successfully developed related products against GLP-1 and GLP-1 receptor (GLP1R) in human and other species. The detailed information is shown in Table 1, these products provide a very sound technical platform for researchers to study the function of GLP1 and its receptors.
