  • Modeling Period4-6 weeks
  • Modeling MethodMale C57BL/6 mice, 4-week old, weight 300 to 400g, no ear infection. The mice were perfused with anesthesia, Sterile with 1mg/L aprotinin in 10mmol/L Tris buffer to isolate all the inner eardrum tissue, homogenate. Homogenate the supernatant. Immunized with prepared inner ear antigen (IEAg) mice, each model mouse was injected subcutaneously in the back of the neck, 100ug each. The mice were treated with auditory brainstem response after immunization. Animal anesthesia after the sound, electric screen spacious room, using evoked potentiometer test. Take the stimulus 0.1ms as short, the interval is 50ms, filter 100 ~ 3 000Hz, scan time is 10ms, superimposed 500 times. Then 30dB below the stimulus, the white noise is masked against the side ear. The reaction valve of ABR is judged by the reaction valve of wave II. The anti-inner ear antigen antibody was detected in animal serum at day 7, 14, 21 and 28 days, and the temporal slices were made from the inner ear tissue. After HE staining, light microscopy.
