MSG1 transcripts in pigmented human epidermal melanocytes but did not detect MSG1 expression in any of the examined depigmented or weakly pigmented human melanoma cells. The deduced 193-amino acid human MSG1 protein shares 75% overall sequence identity with mouse Msg1. The C-terminal 54 amino acids of these proteins are 96% identical and highly acidic, with 22% of the amino acids being either aspartic acid or glutamic acid; this region forms a predicted amphipathic alpha-helical structure. Immunoprecipitation studies using an antibody against Msg1 detected a 27-kD protein in human epidermal melanocyte lysates. Immunofluorescence cytochemistry localized MSG1 to the nucleus of normal human epidermal melanocytes and human melanoma cells.
