Signal-transducing adaptor molecule (STAM) acts downstream of interleukin-2 (IL2)-induced signaling through JAK3. It also interacts with JAK2 after granulocyte-macrophage colony-stimulating factor (GMCSF) stimulation. STAM contains an SH3 domain that is required for induction of MYC and cell growth.
STAMBP binds to the SH3 domain of the signal-transducing adaptor molecule, and plays a critical role in cytokine-mediated signaling for MYC induction and cell cycle progression. Multiple alternatively spliced transcript variants encoding the same protein isoform have been found for this gene.The deduced 424-amino acid protein contains 2 potential SH3-binding domains (PxxP motifs), a JAB1 subdomain homologous (JSH) region, and a putative bipartite nuclear localization signal.
