How to Detect HMGB1 (High Mobility Group Box 1) in-vitro

High Mobility GroupProtein 1, also known as amphotericin. It’s encoded by HMGB1 genes. It’s namedwith HMGB1 due to the high mobility in SDS-PAGE.

Now we treat humanHMGB1 as an example to have a brief introduction on High Mobility Group Protein1.

Protein:

Like Histones, HMGB1is the most important chromatin protein, it plays significant roles inmaintaining and stabilizing nucleosome DNA recombination, replication,transcription, repair etc. In addition, it also functions in infection,inflammation and immune reactions. HMGB1 is also one of the focus ofinflammation injury. Some scholars proposed HMGB1 as a DNA vaccine adjuvant andcancer therapeutic target.

According UniprotMaster HMGB1 link[A1] , http://www.uniprot.org/uniprot/P09429, weprepared three HMGB1 fragment. It’s Met1 ~ Glu215(RPA399Hu01), Pro9 ~ Arg163 (RPA399Hu02),Met1 ~ Glu215(RPA399Hu61).RPA399Hu01 and RPA399Hu01 is expressed in E.coli, and RPA399Hu61 is expressedby 293F. Thus the researchers could choose different products according totheir experimental purposes.


Antibody: Immunizingmice and rabbits with proteins above to get monoclonal and polyclonalantibodies. Then purified with affinity chromatography to get high purityantibodies. The titer of monoclonal antibody MAA399Hu22 could reach 81w.

Kit:

After obtainingmonoclonal and polyclonal antibodies, we developed Elisa kits for HMG1. Kit iscoated with mouse monoclonal antibodies, and detected with rabbit polyclonalantibodies. [A1] The content of HMG1 varies in normal and diseasemodels, the Cloud-Clone. Corp. detected many samples, from 46 apparently healthy people, the concentration isfrom 72 ~ 582ng / ml, for the customer’s reference.
