Library Introduction

cDNA library (Complementary DNA Library) is a mixture of various contructs, which contains different cDNA fragments which is created from a mature mRNA from eukaryotic cells with the use of an enzyme known as reverse transcriptase. Those constructs are easy to express various proteins, unlike genome, therefore target genes can be screened from cDNA library and expressed directly.

By constructing cDNA expression library, the probe of molecular marker linkage map can be provided, more importantly, it can be used to isolate full-length target gene and study its biological function. Thus, cDNA library has special advantage in the study of certain cell expression status of specific genomic and gene function identification. It has a broader application for the research of the individual development, cell differentiation, cell cycle regulation, apoptosis, and other biological phenomena.

Cloud Clone Corp. cDNA library

Construction method:

Cloud Clone cDNA library uses Oligo (dT) as a reverse primer for the synthesis of cDNA , our vector is named Cloud Clone vector (pUSCN) with a unique linker.

Cloud Clone library quality standard: 

1. Inserted cDNA fragment> 700bp;

2. The library capacity (titer) reaches 10^6;

3. Randomly-selected 20 transformed clones are sequenced to verify the cDNA species;

Primer is M13, Lane 13 is 200bp Marker and Lane 24 is 1000bp Marker.

Current library:

General libraries are from human, mouse, rat, pig, cattle and other species tissues;

Now we can supply Level 1 library(cloning plasmid). Customers can use two-hybrid yeast or other suitable experiments to establish Level two library.

Custom Service

Service:

1. RNA extraction: Total RNA extraction, isolation of mRNA;

2. PCR synthesis: first strand synthesis, connectors, second strand synthesis and amplification;

3. Screening fragments: select the length of double-strand cDNA;

4. vector: ligation reaction of the selected cDNA and pUSCN vector ;

5. Transformation: transfrom vector into competent cells;

6. Quality control: cDNA library storage capacity detection, each library randomly selected 20 clones to detect insert size by PCR.
